{"id":420,"date":"2014-07-11T11:07:32","date_gmt":"2014-07-11T15:07:32","guid":{"rendered":"http:\/\/blogs.shu.edu\/cancer\/?p=420"},"modified":"2021-07-02T08:51:59","modified_gmt":"2021-07-02T12:51:59","slug":"novel-mutated-antigens-from-tumor-infiltrating-lymphocytes-from-long-term-melanoma-responders","status":"publish","type":"post","link":"https:\/\/blogs.shu.edu\/cancer\/2014\/07\/11\/novel-mutated-antigens-from-tumor-infiltrating-lymphocytes-from-long-term-melanoma-responders\/","title":{"rendered":"Novel mutated antigens from Tumor Infiltrating Lymphocytes from long-term melanoma responders"},"content":{"rendered":"<p>Tumor Infiltrating Lymphocytes (TILs) are cytotoxic T-cells that are found in tumors.\u00a0 They serve to destroy cancer, or at least keep cancer at bay.\u00a0 Adoptive immunotherapy,<!--more--> in which TILs are isolated from patients\u2019 tumors, expanded <em>ex vivo <\/em>via incubation with IL-2, and re-infused autologously, has shown an objective regression rate of 72% in multiple clinical trials of metastatic malignant melanoma (see \u00a0<a title=\"Efficient Identification of Mutated Cancer Antigens Recognized by T Cells Associated with Durable Tumor Regressions\" href=\"http:\/\/clincancerres.aacrjournals.org\/content\/20\/13\/3401.abstract?sf27992416=1\" target=\"_blank\" rel=\"noopener\">http:\/\/clincancerres.aacrjournals.org\/content\/20\/13\/3401.abstract?sf27992416=1<\/a> and <a title=\"Expansion of human tumor infiltrating lymphocytes for use in immunotherapy trials\" href=\"http:\/\/www.sciencedirect.com\/science\/article\/pii\/S0022175987800182\" target=\"_blank\" rel=\"noopener\">http:\/\/www.sciencedirect.com\/science\/article\/pii\/S0022175987800182<\/a>).<\/p>\n<p>Determining the precise cancer antigens to which the TILs are reactive has been elusive.\u00a0 Two patients (2359 and 2591) had complete regression of melanoma metastases in excess of 5 years with TIL therapy.\u00a0 By screening tandem mini-gene libraries, identification of mutated kinesin family member 2C (KIF2C) antigen as a target of TIL 2359, and mutated DNA polymerase alpha subunit B (POLA2) antigen as a target of TIL 2591.
