{"id":283,"date":"2014-06-26T10:44:35","date_gmt":"2014-06-26T14:44:35","guid":{"rendered":"http:\/\/blogs.shu.edu\/cancer\/?p=283"},"modified":"2021-07-02T08:52:00","modified_gmt":"2021-07-02T12:52:00","slug":"aspirin-reduces-risk-of-pancreatic-cancer-by-48","status":"publish","type":"post","link":"https:\/\/blogs.shu.edu\/cancer\/2014\/06\/26\/aspirin-reduces-risk-of-pancreatic-cancer-by-48\/","title":{"rendered":"Aspirin reduces risk of pancreatic cancer by 48%"},"content":{"rendered":"<p>Aspirin was already known to reduce the likelihood of developing colon, esophageal, lung, and prostate cancers. \u00a0Now, add pancreatic cancer to the list&#8230;<!--more--><\/p>\n<p>The mechanism by which low dose chronic aspirin therapy works is by reducing inflammation. \u00a0Chronic inflammation is a cancer promoter, that is, it sets conditions necessary for mutations to occur (proliferation) and establishes the environment for heterotypic cellular interactions with inflammatory cells that help to induce the cancer phenotype and then drive cancer development.<\/p>\n<p>Inflammatory cells secrete TNF (tumor necrosis factor), which induces protein kinase C (PKC), which induces AP-1 and cyclin D-1 leading to proliferation and p53 blockade via mdm2 induction. \u00a0TNF directly induces NFkB, which is also indirectly induced by PKC. \u00a0NFkB is a real bad actor, inducing inhibitors of apoptosis, cyclin D, and myc, which induces telomerase, leading to immortalization. \u00a0As if that weren&#8217;t enough, NFkB also induces COX-2, which induces Prostaglanding E-2. \u00a0PGE2 promotes EMT, loss of contact inhibition, more proliferation, and blocks apoptosis.
