{"id":2763,"date":"2015-12-09T11:05:16","date_gmt":"2015-12-09T16:05:16","guid":{"rendered":"http:\/\/blogs.shu.edu\/cancer\/?p=2763"},"modified":"2021-07-02T08:51:50","modified_gmt":"2021-07-02T12:51:50","slug":"second-multiple-myeloma-antibody-approved-empliciti","status":"publish","type":"post","link":"http:\/\/blogs.shu.edu\/cancer\/2015\/12\/09\/second-multiple-myeloma-antibody-approved-empliciti\/","title":{"rendered":"Second Multiple Myeloma Antibody Approved \u2013 Empliciti"},"content":{"rendered":"<p>Elotuzumab (Empliciti) is now the <a href=\"http:\/\/www.fda.gov\/NewsEvents\/Newsroom\/PressAnnouncements\/ucm474684.htm\" target=\"_blank\" rel=\"noopener\">second monoclonal antibody (Mab) approved by the FDA for multiple myeloma (MM)<\/a>. <a href=\"http:\/\/blogs.shu.edu\/cancer\/2015\/11\/25\/darzalex-for-refractory-multiple-myeloma\/\" target=\"_blank\" rel=\"noopener\">Darzalex (daratumumab),<\/a> the first Mab for MM, was approved by the FDA just 3 weeks ago.<!--more--><\/p>\n<p>The <a href=\"http:\/\/www.wsj.com\/articles\/fda-approves-new-blood-cancer-drug-empliciti-1448901505\" target=\"_blank\" rel=\"noopener\">FDA approved Empliciti<\/a> to be used in combination with two other FDA-approved treatments, dexamethasone and Celgene Corp.\u2019s Revlimid. In a 646-participant clinical trial, those treated with the three-drug cocktail took longer to relapse and had a higher chance of tumor shrinkage than those treated with the two-drug therapy.<\/p>\n<p style=\"padding-left: 30px\">The safety and efficacy of Empliciti were tested in a randomized, open-label clinical study of 646 participants whose multiple myeloma came back after, or did not respond to previous treatment. Those taking Empliciti plus Revlimid and dexamethasone experienced a delay in the amount of time before their disease worsened (19.4 months) compared to participants taking only Revlimid and dexamethasone (14.9 months). Additionally, 78.5 percent of those taking Empliciti with Revlimid and dexamethasone saw a complete or partial shrinkage of their tumors compared to 65.5 percent in those only taking Revlimid and dexamethasone.<\/p>\n<p>Multiple myeloma is a type of blood cancer that occurs white blood cells found in the bone marrow. The National Cancer Institute estimates there will be 26,850 new multiple myeloma cases and 11,240 related deaths in the U.S. this year. Empliciti activates the body\u2019s own immune system to attack and destroy multiple myeloma cells.<\/p>\n<p><a href=\"http:\/\/lmt.projectsinknowledge.com\/Activity\/index.cfm?showfile=b&amp;i=26&amp;jn=2154&amp;sj=2154.31\" target=\"_blank\" rel=\"noopener\">Multiple myeloma may be preceded<\/a> by either monoclonal gammopathy of undetermined significance (MGUS) or smoldering multiple myeloma (SMM). MGUS is a premalignant condition that is associated with a 1% per year progression to multiple myeloma. Smoldering multiple myeloma (SMM) resembles MGUS in the absence of end-organ involvement, but is much more likely to progress to myeloma.<\/p>\n<p><a href=\"http:\/\/blogs.shu.edu\/cancer\/files\/2015\/12\/MM-clinical-diagnosis.gif\" data-rel=\"lightbox-image-0\" data-rl_title=\"\" data-rl_caption=\"\" title=\"\"><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter size-full wp-image-2766\" src=\"http:\/\/blogs.shu.edu\/cancer\/files\/2015\/12\/MM-clinical-diagnosis.gif\" alt=\"MM clinical diagnosis\" width=\"500\" height=\"386\" \/><\/a><\/p>\n<p>Elotuzumab is a <a href=\"http:\/\/blogs.shu.edu\/cancer\/?p=1882\" target=\"_blank\" rel=\"noopener\">humanized monoclonal antibody that binds to CS1 (SLAMF7)<\/a>, which is expressed on MM cells, as well as NK cells. This new mechanism of action is very important for those patients who don\u2019t have other options.
